One of the best (maybe it IS the best) bookstores in Vancouver, for both used and new books. They heavily discount new books, they can get just about anything, and they turn most orders around very quickly. Don't spend your money with Amazon - support local businesses, like this one!

My favourite indie bookstore. Staff is fantastic and very knowledgeable. Prices are great. Selection is great. Can't say enough good things about it.

This main street location has a great used books section in the back. It's much bigger than it looks from the outside.

Solid bookstore. Well organized compared to other local used shops. Good selection. Awesome hours. 
Be warned though that the proprietor gives of a distinct Comic Book Guy vibe. I've overheard him trashing customers after leaving a couple times, both were people who tried to sell him books. He can come off as pretty adversarial in this regard. 
You can order books from them for amazon prices or less, but they may come quite a bit later than you expect and there's no tracking. For new books I prefer Book Warehouse if looking to shop local.
